جداسازی و شناسایی باکتری‌های لجن مازاد فاضلاب شهری و بررسی مقاومت آن‌ها در برابر آموکسی سیلین و جنتامایسین

چکیده
زمینه و هدف: مدیریت پایدار لجن فاضلاب شهری (MSS) و تثبیت آن به‌منظور کاهش پاتوژن‌ها و بوهای نامطبوع، از ضرورت‌های زیست‌محیطی حائز اهمیت است. با این‌حال، ورود باقی‌مانده‌های آنتی‌بیوتیکی به سیستم‌های فاضلاب می‌تواند بر تنوع و عملکرد جوامع میکروبی تأثیر گذاشته و چالش‌های جدیدی را در فرآیندهای تثبیت بیولوژیکی ایجاد کند. بر این اساس، پژوهش حاضر با هدف جداسازی و شناسایی سویه‌های باکتریایی از نمونه‌های لجن مازاد فاضلاب شهری و بررسی الگوی مقاومت آن‌ها در برابر آنتی‌بیوتیک‌های آموکسیسیلین و جنتامایسین طراحی گردید تا میزان اثرگذاری این ترکیبات بر عملکرد این سویه‌ها مورد ارزیابی قرار گیرد.

مواد و روش‌ها: در این مطالعه توصیفی-تحلیلی، سویه‌های باکتریایی از لجن مازاد فاضلاب شهری جداسازی شدند. تعیین هویت این سویه‌ها از طریق آزمون‌های بیوشیمیایی صورت گرفت و الگوی مقاومت آنتی‌بیوتیکی آن‌ها در برابر دو آنتی‌بیوتیک آموکسیسیلین و جنتامایسین، با استفاده از روش انتشار دیسک (Disk Diffusion) مورد ارزیابی قرار گرفت.

یافته‌ها: نتایج بررسی‌ها نشان داد که میزان مقاومت آنتی‌بیوتیکی در عوامل باکتریایی جداشده از لجن مازاد به‌طور قابل‌توجهی بالا است. در میان آنتی‌بیوتیک‌های بررسی شده، بیشترین میزان مقاومت مربوط به آموکسیسیلین بود.

نتیجه‌گیری: یافته‌های این پژوهش حاکی از آن است که مصرف بی‌رویه و نادرست آنتی‌بیوتیک‌ها در بخش بهداشت و درمان، منجر به افزایش مقاومت میکروبی در محیط‌های تصفیه فاضلاب شده است. لذا بازنگری در پروتکل‌های مدیریت داروهای مصرف‌نشده و بهینه‌سازی روش‌های تثبیت لجن با در نظر گرفتن اثرات آنتی‌بیوتیک‌ها توصیه می‌گردد.

Background and Objective: Sustainable Municipal Sewage Sludge Management (MSS ) and its stabilization to reduce pathogens and unpleasant odors are critical environmental imperatives. However, the influx of antibiotic residues into wastewater systems can affect the diversity and function of microbial communities, posing new challenges for biological stabilization processes. Consequently, the present study was designed to isolate and identify bacterial strains from municipal excess sludge samples and analyze their resistance patterns against amoxicillin and gentamicin, in order to evaluate the impact of these compounds on the performance of these strains.

Materials and Methods: In this descriptive-analytical study, bacterial strains were isolated from municipal waste activated sludge. The strains were identified via biochemical tests, and their antibiotic resistance patterns against amoxicillin and gentamicin were evaluated using the disk diffusion method.

Results: The results demonstrated a significantly high level of antibiotic resistance among the bacterial agents isolated from the waste activated sludge. Among the antibiotics studied, the highest resistance was observed toward amoxicillin.

Conclusion: The findings indicate that the excessive and improper use of antibiotics in healthcare has led to increased microbial resistance within sewage treatment environments. Consequently, it is recommended to revise protocols for managing unused medications and optimize sludge stabilization methods by considering the influence of antibiotics.
