مقاومت آنتی‌بیوتیکی و تایپینگ ژن agr در جدایه‌های استافیلوکوکوس‌اورئوس از گوشت مرغ و سالادهای آماده مصرف در هرمزگان، ایران

چکیده
زمینه و هدف: استافیلوکوکوس اورئوس، به‌عنوان یکی از چالش‌برانگیزترین پاتوژن‌های منتقله از غذا، توانایی بالایی در کسب ژن‌های مقاومت آنتی‌بیوتیکی دارد. پژوهش حاضر با هدف واکاوی میزان تطابق الگوی مقاومت فنوتیپی و ژنوتیپی و هم‌چنین تعیین تنوع سیستم تنظیمی agr در جدایه‌های به‌دست آمده از گوشت مرغ و سالادهای الویه و سزار در هرمزگان به مرحله‌ی اجرا درآمد.

مواد و روش‌ها: در این پژوهش مقطعی، ۱۳۰ نمونه شامل گوشت مرغ (75=N) سالاد الویه (30=N) و سالاد سزار (25=N) بررسی شد. پس از جداسازی باکتریولوژیک و تایید گونه، حساسیت فنوتیپی ۴۸ جدایه نسبت به ۱۱ آنتی‌بیوتیک سنجیده شد. سپس حضور ۱۱ ژن مقاومت (شامل ermA/C، msrA/B، tetK/M، aacA-D، vatA/B/C و linA) و تنوع تیپ‌های agr به روش PCR  مورد ارزیابی قرار گرفت.

یافته‌ها: از مجموع نمونه‌ها، ۴۸ مورد (36/92%) آلوده به استافیلوکوکوس اورئوس بودند که بیشترین میزان آلودگی در سالاد الویه (56/66%) مشاهده شد. در بررسی فنوتیپی، بیشترین مقاومت نسبت به آزیترومایسین و کلیندامایسین ثبت گردید. نتایج مولکولی نشان داد که ژنermC  با فراوانی بالا (تقریباً در تمامی سویه‌ها) غالب‌ترین ژن مقاومت است. هم‌چنین، در تایپینگ سیستم تنظیمی، تیپ agr IV با 35/4% (۱۷ مورد) بیشترین فراوانی را داشت و پس از آن تیپ‌هایagr II  (%27) وagr I  (20/8%) قرار گرفتند.

نتیجه‌گیری: نتایج این مطالعه نشان‌دهنده‌ی بار میکروبی و شیوع بالای سویه‌های MDR با غلبه ژنتیکی تیپagr IV  در غذاهای آماده مصرف است. یافته‌ها ضرورت نظارت دقیق‌تر بر بهداشت پرسنل و زنجیره فرآوری سالادهای آماده مصرف را به‌منظور کاهش ریسک انتقال باکتری‌های مقاوم به مصرف‌کنندگان برجسته می‌سازد.

Antibiotic Resistance and agr Typing of Staphylococcus aureus Isolates from Ready-to-Eat Chicken Meat and Salads in Hormozgan, Iran

Background and Objective: Staphylococcus aureus is a significant foodborne pathogen known for its extensive antibiotic resistance capabilities. This study aimed to evaluate the correlation between phenotypic and genotypic resistance patterns and determine the diversity of the accessory gene regulator (agr) system in isolates recovered from chicken meat and cold salads (Olivier and Caesar) in Hormozgan, Iran.

Materials and Methods: In this cross-sectional study, 130 samples including chicken meat (n=75), Olivier salad (n=30), and Caesar salad (n=25) were analyzed. Following bacteriological isolation and species confirmation, the phenotypic susceptibility of 48 isolates was tested against 11 antibiotics. The presence of 11 resistance genes (including ermA/C, msrA/B, tetK/M, aacA-D, vatA/B/C, and linA) and agr types were evaluated using conventional PCR.

Results: Out of 130 samples, 48 (36.92%) were positive for S. aureus, with the highest contamination rate found in Olivier salad (56.66%). Phenotypically, the highest resistance was observed against Azithromycin and Clindamycin. Molecular results revealed that the ermC gene was the most prevalent resistance marker. Regarding the regulatory system, agr IV was the predominant genetic profile (35.4%, 17 cases), followed by agr II (27%) and agr I (20.8%).

Conclusion: The findings indicate a high microbial load and an alarming prevalence of MDR strains dominated by the agr IV genotype in RTE foods. These results emphasize the need for stricter monitoring of personnel hygiene and the processing chain of RTE salads to mitigate the risk of transmitting resistant bacteria to consumers.
